A workhorse of the West Side, route 68 Guadalupe provides frequent service along Frio Street and Guadalupe Street. The route continues on Old Castroville Road to make a stop at the Las Palmas Shopping Center, and then every other trip continues on to 36th Street before turning around to head back downtown.

The fifteen-minute frequencies on route 68 provide a high level of service to such locations as the University Family Health Center, the Guadalupe Cultural Arts Center, and the Texas Diabetes Institute, which is a part of the University Health System. The Diabetes Institute is on Zarzamora Street just one block away from route 68, and it has the distinction of being the nation's largest diabetes care center. Established in 1993, the institute is a research center that also provides basic care, patient education, treatment, and professional training.
